Two determinants of symptom pattern
Pathway reliance: how much a tissue depends on the blocked pathway. Cell turnover rate: how quickly a tissue replaces its cells. A tissue that is both highly reliant and fast-turnover will show symptoms earliest.

A block in a pathway needed for DNA synthesis affects bone marrow and gut lining first because those cells divide rapidly. A block in a pathway needed for myelin maintenance affects nerves, which turn over slowly, so neurological symptoms may appear later or after other signs.
Clinical implication
The order in which symptoms appear is not random. It reflects which tissues are most dependent on the blocked pathway and how quickly those tissues renew. This is why early deficiency signs often involve rapidly dividing tissues.
